Abstract

An investigation of the fresh leaves of Mitragyna speciosa has resulted in the isolation of a new alkaloid in addition to the indole alkaloids previously reported. The new alkaloid is the 3-dehydro derivative of mitragynine and its structure was elucidated by spectral means and chemical transformations. (-)-Epicatechin was also isolated from the leaves.

Mitragyna speciosa Korth. leaves have been used for decades as a traditional medicine to treat diarrhea, diabetes and to improve blood circulation by natives of Malaysia, Thailand and other regions of Southeast Asia. Mitragynine is the major active alkaloid in the plant. To date, the role of mitragynine in psychological disorders such as depression is not scientifically evaluated.

Abstract
Acute administration of Mitragyna speciosa (MS) extract (45 and 50 mg/kg) significantly resulted in dose-dependent decreases in food and water intakes (P < 0.05) in rats. Prolonged suppressing effects were observed following administration of the MS extract (40 mg/kg) for 60 consecutive days. Moreover, the long-term administration also significantly suppressed weight gaining.

Abstract
7-Hydroxymitragynine is a potent opioid analgesic alkaloid isolated from the Thai medicinal herb Mitragyna speciosa. In the present study, we investigated the opioid receptor subtype responsible for the analgesic effect of this compound. In addition, we tested whether development of tolerance, cross-tolerance to morphine and naloxone-induced withdrawal signs were observed in chronically 7-hydroxymitragynine-treated mice.

Ethnopharmacological relevance: Mitragyna speciosa Korth (ketum) is widely used in Malaysia as a medicinal agent for treating diarrhea, worm infestations and also acts as an analgesic and antipyretic.

The aim of the study is to determine the acute toxicity of Mitragyna speciosa Korth standardized methanol extract in vivo in 4-weeks-old Sprague–Dawley rats. Result No mortality was noted after 14 days of treatment. In general, behavior, food and water consumption, hematological studies and organ weights showed no significant changes.

In the present study, we investigated the opioid receptor subtype responsible for the analgesic effect of this compound. In addition, we tested whether development of tolerance, cross-tolerance to morphine and naloxone-induced withdrawal signs were observed in chronically 7-hydroxymitragynine-treated mice. Subcutaneous (s.c.) administration of 7-hydroxymitragynine produced a potent antinociceptive effect mainly through activation of μ-opioid receptors.
